          Sudan Black Stain

          Sudan Black Stain

           

          Intended use:

          Sudan Black stain is intended for neutral fat staining.

           

          Principle:

          It is thought that the staining of lipids depends on the physical properties of solution or adsorption. When the Sudan Black stain is dissolved in organic solvent, it exhibits a greater solubility in lipoids of frozen tissue than that in original solvents. So during staining, dyes will migrate into lipids from organic solvents resulting in lipid staining.

           

          Methods:

          1. Cut frozen tissue at 6~10μm, rinse in distilled water.

          2. Rinse tissue in 50~70% ethanol for 20~30 seconds.

          3. Place in Sudan Black B solution for 3~5 minutes.

          4. Differentiate in 50~70% ethanol for a few seconds until the loose color on slides is removed.

          5. Rinse in distilled water.

          6. Counterstain in Nuclear Fast Red solution for 5 minutes.

          7. Rinse in distilled water for 1~2 minutes.

          8. Remove any excess water around the slide then dry in the air.

          9. Mount with arabinan or glycerin jelly.

          Precaution:

          1. To display lipids, stain frozen tissue, instead of paraffin tissue, as lipids are readily soluble in organic solvents.

          2. The frozen tissue used for lipid staining should not be too thin. Otherwise, the lipid content may be lost.

          3. The stain will not remain for a long time, so observe and take pictures as soon as possible.
